Deletions or editorial revisions made to these sections are not included in this summary. Read about the new physician labeling format. Summary View Sections Modified PRECAUTIONS Drug Interactions Drug Metabolized by CYP2D6 Therefore, coadministration of Paxil CR with other drugs that are metabolized by this isozyme, including certain drugs effective in the treatment of major depressive disorder (e.g, nortriptyline, amitriptyline, imipramine, desipramine, and fluoxetine ), phenothiazines, risperidone, tamoxifen, and Type 1C antiarrhythmics (e.g, propafenone, flecainide, and encainide ), or that inhibit this enzyme (e.g, quinidine ), should be approached with caution .Tamoxifen is a pro-drug requiring metabolic activation by CYP2D6. Inhibition of CYP2D6 by paroxetine may lead to reduced plasma concentrations of an active metabolite and hence reduced efficacy of tamoxifen.-- Links on this page: Page Last Updated: 06 / 19 / 2009 Note: If you need help accessing information in different file formats, see Instructions for Downloading Viewers and Players. ... 3.8 %in hip BMD compared to a median decrease of 2.0 %in the placebo group. The changes from baseline in lumbar spine BMD in letrozole and placebo treated groups were not statistically different. Cholesterol Consideration should be given to monitoring serum cholesterol. In the adjuvant setting, an increase of â ‰¥ 1.5 X ULN in total cholesterol (generally non-fasting) was observed in patients who had baseline total serum cholesterol within the normal range in 151 / 1843 (8.2 %) on letrozole vs 57 / 1840 (3.2 %) on tamoxifen. Lipid lowering medications were required for 25 %of patients on letrozole and 16 %on tamoxifen. ADVERSE REACTIONS Updated Analysis, Extended Adjuvant Treatment of Early Breast Cancer, Median Treatment Duration of 60 Months The extended adjuvant treatment trial was unblinded early. At the updated (final analysis ), overall the side effects seen were consistent to those seen at a median treatment duration of 24 months.-- Links on this page: Page Last Updated: 04 / 15 / 2010 Note: If you need help accessing information in different file formats ...
